Clomiphene is a weak estrogen. The estrogenic effects are best demonstrated in animals with marked gonadal deficiency. Clomiphene has also been shown to effectively inhibit the action of stronger estrogens. In humans, ¡t leads to an increase in the secretion of gonadotrophins and estrogens. The pharmacological importance of this compound rests on its ability to stimulate ovulation in women with amenorrhea and other ovulatory disorders. The mechanism by which ovulation ¡s produced is not known. It has been suggested that it blocks as inhibitory influence of estrogen on the hypothalamus and increases the production of gonadotrophins Clomiphene will not work in the absence of a pituitary gland capable of producing gonadotrophinsi. It has been shown ¡n the rat that crystals of this compound implanted in tile pituitary have no effect, whereas those implanted in parts of the median eminence are ale to induce ovulation. The observation that this compound can induce ovulation in patients with very low estrogen levels does not support the hypothesis that it works by breaking a feedback inhibition. However, it does stimulate gonadotrophin production by the pituitary byh an action on the hypothalamus.
